Description

Description: This assay employs a two-site sandwich ELISA to quantitate HIST1H2AG in samples. An antibody specific for HIST1H2AG has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HIST1H2AG present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HIST1H2AG is added to the wells. After washing, Streptavidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HIST1H2AG bound in the initial step. The color development is stopped and the intensity of the color is measured.
Overview: Histones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Two molecules of each of the four core histones form an octamer, around which approximately 146 bp of DNA is wrapped in repeating units, called nucleosomes. The linker histone, H1, interacts with linker DNA between nucleosomes and functions in the compaction of chromatin into higher order structures.
